I was being asked so much how I color my art.
Since I'm not Painter professional user ;moreover, I often change and find new tehcnic so I'm not sure how to make a tutorial.
Anyway I have 2-3 tools and 2-3 technics I usually use while painting in Painter 9.
This is the one how I draw and color.
It's not the best way and it's fit for someone who already know some tools in Painter (I'm lazy to point out where is anything)
If my English doesn't make sense, you may ask your question here. I'll try my best to answer.

Edit - About the Digital Water Color, I used Simple Water.

It cause the speed of monitor displaying of any program even they're not game or 3D program, though you didnt see it obviously .
If you used water color in Painter, you'll know that there are animations in the program.
